Is it possible as an instructor to take a student's discussion reply that was not a reply to the original post, but rather to another student's reply, and disassociate it from the "parent"?
I know the "must post to see peer's responses", but even with that, the functionality does not exist for the subsequent post from a student to be isolated as an original response.

Canvas does not support moving a post to a different parent.
Aside from the post first option you mentioned, you can ask the student to post it again in the correct location and then delete (or edit) the original.
