My partner teacher & I shared Sandbox Courses for each of our preps. She is leaving the district. When her employment officially ends and her account is deactivated, will I lose access to the Sandbox Course she initially created? Or will I still have access since I am an Instructor in the course, regardless of who created it?

The course will not disappear when her account is removed, just her access to the course is removed. If you were enrolled in that course as a teacher, then you will still have access to that course and any content within it.
