How can an instructor see if a student has read a comment left for them.

If you are talking about annotations on an assignment submission, you can see when the student viewed the graded assignment: https://community.canvaslms.com/docs/DOC-6092#jive_content_id_SpeedGrader
Other than asking the student to tell you when they view a submission comment on a discussion or quiz or assignment, the indicator I referenced above is the only way.

I'm not clear on what part of Canvas the student would be reading a comment from the instructor. Would that be as part of a submitted assignment via the SpeedGrader? As a discussion posting? As an instructor you can go to the "People" page in your course and then click on the "Student Interactions Report" button on the right side of your screen which will give you some information. And, you can also click on the cog wheel next to a student's name and then select "User Details". From this screen, select "Access Report for [insert name]". I'm not sure whether these screens will get you the exact information you are looking for, but it might be a start at least.
