Hi, I have to create an Assignment and grade it.. 

Then I have to use the annotation tab to "mark up"??

Where is the tab and what do I do exactly..

Hello  @ostranderi ...

The SpeedGrader in your Canvas course does not have a button called "Annotations", but it does have an annotations toolbar.  This is briefly discussed just before half-way through this short video tutorial: .  You can use the annotation tools to annotate over a student submission in the SpeedGrader.  This written Guide will also be of help to you: How do I add annotated comments in student submissions using DocViewer in SpeedGrader?.
