Good Afternoon,
I was hoping to have some assistance or advice regarding auto-responses in the Canvas Inbox.
The setting is enabled. I have created an out-of-office time frame and message as well as turned it on. The characters of the email are less than 255. However, when students or anyone else responds to my emails, the auto-response will not be delivered. Additionally, if anyone emails me first, the auto-response will not work either.

I don't have a solution for you, but my supervisor and I were testing out this same thing last week, and he never received an out-of-office response, either. You might want to report this to Canvas Support so they can take a look at it and address any potential issues.

I just tested, and it's not working for me either. I guess it's just another of the recently added features that I have to disable because it's not working.
We tested it on our Dev instance and it worked fine. When we flipped it on in Production and tested, suddenly it did not work, so we did a full-stop and turned it back off. Does anyone have an active ticket we can request to be added to? L2 is asking for screenrecordings, etc (basically for us to test it all over again), and I would really rather not! 🙂

Just tagging along as well - it seems to work intermittently. I tested with a colleague, and worked great. I tested in a live course w/ a Prof who reported it wasn't working and it didnt work. I disabled auto-reply, and re-enabled, tested and it worked. I gave Prof the Green light, just to come back and report it doesn't work anymore.
I'll be opening a ticket soon.
