Can an instructor lock an assignment portal after the assignment is graded?

I am an instructor in multiple courses. Students are submitting to an assignment portal after the assignment has already been graded. My question is if the assignment portal can be set to "locked" after a grade is given to a submitted assignment to avoid additional submissions.

Do you mean that students are submitting a first attempt after you start grading the assignments, or do you mean that students are resubmitting the assignment after you graded their first attempt.

If students are resubmitting assignments after the assignment was due, change "Submission Attempts" to "Limited" and set the "Number of Attempts" to "1."

I assume this assignment was already been due before You graded it. What is the "Until" date set to? I set it to the last date and time I want to students to be able to submit that assignment. If you change the "Until" date and time, nobody will be able to submit the assignment after that. I wait until my "Until" date and time have passed to grade assignments.

Students are submitting the assignment, I grade it and then they submit another assignment over it that belongs in Assignment 2. That second assignment is an update that incorporated the feedback I provided but the instructions are to submit the "revised" document to Assignment 2 for the next grade. 

The attempts can't be set to limited because they need a 60 to pass Assignment 1. They can receive either a "0" meaning they did not meet the requirement or a " 60" meaning the benchmark was met. If they get a zero, they have to be able to resubmit so they can pass it and then submit a revised version to Assignment 2 for the balance of the points needed to pass. There are 100 - 125 students in each class so we can not wait until the last day to grade.
