When I sync my grades, only 5 out of the 6 assignments I have on Canvas are syncing over to my gradebook. I have ensured that the assignment not syncing is under the same categories in both Canvas and my gradebook and has a due date. I have also tried remaster the gradebook and that assignment continues to be left out. What am I missing?

Have you tried looking at the data from the Grade Sync tab on your course navigation menu? I don't know where there is documentation about it, and I don't understand it very well. But often I can find an error message there that shows me why a certain assignment and/or course did not sync properly.

I did check and I am not getting an error message. I re-synced and now it went through. I am not sure what the mistake was as I did not change anything. I did have to sync it 3 times which can be annoying. I caught it now since I have very few assignments, but it makes me nervous for the remainder of the semester.

How long did you wait to post after attempting to sync? Sometimes it just takes a while (up to 24 hours), for no apparent reason. It's not instantaneous.
I usually recommend syncing only a few assignments at a time (at most) because there are so many ways it can go awry, from temporarily dropped (or weak) wi-fi to a minor issue with the other system (your gradebook program) to many other "gremlins." Sticking to 1-3 assignments at a time makes it easier to track progress and catch problems. But I wouldn't worry about the rest of the semester in general. It seems that your process is basically working, with some kind of minor hiccup that occurred this time.
Did that assignment that didn't go through the first time have grades for ALL students?
If it didn't then that might explain the lack of an error and the lack of those grades being pushed through.
